Eigenschaften
Es dient als Vektor und erlaubt im Gegensatz zu den Cosmiden eine Klonierung von grΓΆΓeren Genomabschnitten. Das erste YAC entstand als Kombination eines ARS und eines Zentromers der Hefe mit den Telomeren von Tetrahymena, wodurch ein lineares MolekΓΌl entstand, welches sich in einer Hefe wie ein normales Chromosom verhΓ€lt. Das erste YAC wurde von Murray und Szostak 1983 entwickelt.cite-ref-1[1]
Trotz dieses Vorteils haben die YACs einen entscheidenden Nachteil. Aufgrund ihrer LΓ€nge ist es sehr schwierig, sie in flΓΌssigem Medium zu handhaben und intakt zu halten. Aus diesem Grund versuchte man frΓΌher, die Abschnitte nicht lΓ€nger als 50 β 100 kbp zu konstruieren. Durch eine Fraktionierung per Pulsed-Field-Gelelektrophorese wurden kurze DNA-Fragmente aus dem MolekΓΌl entfernt und die durchschnittliche LΓ€nge konnte auf 350 kbp verlΓ€ngert werden.cite-ref-2[2] Durch den Einsatz von Polyaminen wurde eine Stabilisierung der MolekΓΌle erreicht, wodurch zum Einsatz beim Mensch- und beim Mausgenom Insertionen von bis zu 700 kbp mΓΆglich wurden.cite-ref-3[3] 1992 wurde eine Genbibliothek mit durchschnittlichen InsertionsgrΓΆΓen von 810 kbp entwickelt, wobei einige MolekΓΌle LΓ€ngen von bis zu 1800 kbp erreichten.cite-ref-4[4]
